The kaolin clotting time; a rapid one-stage method for diagnosis of coagulation defects.

چکیده

A major variable in blood coagulation tests is the nature and the amount of surface with which the blood comes in contact. When tests on recalcified plasma are used the methods of preparation and storage of the plasma have also to be considered. The prolonged contact of plasma with a large surface, e.g., with glass powder or asbestos, is accompanied by the loss of many coagulation factors, including prothrombin and fibrinogen (Tocantins, 1945). Even a more limited exposure to glass, such as is likely to be encountered in normal practice, may be followed by considerable prolongation of the plasma clotting time (Margolis, 1957). The loss of clotting ability under these conditions is primarily due to the disappearance of the plasma thromboplastin antecedent (Rosenthal, 1955) which is the component most easily destroyed by contact with glass (Margolis, 1958). An additional difficulty when dealing with plasma is that, as the number of platelets decreases with increasing centrifugation, the clotting time becomes more and more sensitive to variations in the platelet count and the results become more unpredictable. Previous investigations (Margolis, 1957) indicated that the clotting time of re-calcified plasma can yield both sensitive and reproducible results when both the contact surface and the platelet content are controlled. The purpose of the present paper is to demonstrate the usefulness of this procedure for the diagnosis of plasma clotting defects.

Lupus anticoagulants: improved diagnosis with a kaolin clotting time using rabbit brain phospholipid in standard and high concentrations.

We utilized a kaolin-activated partial thromboplastin time (APTT) using rabbit brain phospholipid, in which the capacity of a fourfold increased "high" phospholipid concentration (PC) to normalize the abnormal "standard" PC-APTT in patients with lupus anticoagulants is assessed. This system was also used to measure factors VIIIC, IX, and XI. The tissue thromboplastin inhibition test (TTI), a pr...

Properties of sulfatides in factor-XII-dependent contact activation.

Incubation of normal human plasma with low amounts of sulfatides resulted in the initiation of intrinsic coagulation and the appearance of kallikrein activity. The optimal initiation of procoagulant and kallikrein amidolytic activity was dependent on the presence of factor XII, high molecular weight kininogen, and prekallikrein. Since the activated partial thromboplastin clotting times in preka...

A hitherto undescribed plasma factor acting at the contact phase of blood coagulation (Flaujeac factor): case report and coagulation studies.

This paper reports an asymptomatic coagulation defect responsible for an abnormality at the contact phase of blood coagulation in vitro, distinct from Hageman factor and Fletcher factor deficiencies. Coagulation studies in a 50-yr-old French woman without bleeding tendency revealed the following results: whole-blood clotting time in glass tubes and activated partial thromboplastin time with kao...

Contact activation of blood coagulation on a defined kaolin/collagen surface in a microfluidic assay.

Generation of active Factor XII (FXIIa) triggers blood clotting on artificial surfaces and may also enhance intravascular thrombosis. We developed a patterned kaolin (0 to 0.3 pg/μm(2))/type 1 collagen fibril surface for controlled microfluidic clotting assays. Perfusion of whole blood (treated only with a low level of 4 μg/mL of the XIIa inhibitor, corn trypsin inhibitor) drove platelet deposi...
